Project Completion Report of Dr. Thwe Thwe Win, Myanmar

Dr. Thwe Thwe Win, Assistant Lecturer at Mandalay Technological University of Myanmar was affiliated with the International Centre for Chemical and Biological Sciences (ICCBS), Karachi, Pakistan from 1st September 2009 to 28th February 2010 as a Post-Doctoral Fellow under the Joint NAM S&T Centre - ICCBS Fellowship scheme for the year 2009. She worked on 'Molecular Mechanism of Known and Unknown Chemotherapeutic Agents on Human Carcinoma Cell Lines' under the supervision of Dr. Huma Rasheed, Assistant Professor in the the Panjwani Center for Molecular Medicine and Drug Research (PCMD) of ICCBS. She learned cell culture techniques and performed all procedures from cell seeding to cryo-preservation under the required culture conditions. She treated the cells with various concentrations of drugs and determined the viability and proliferation of the cells using trypan blue exclusion assay. The cytotoxicity study was performed by fluorimetric assay at different time intervals using spectrofluorometer and IC50 values of the compounds were determined. She also gained experience of isolating DNA with Trizol Reagent for DNA fragmentation using agarose gel electrophoresis; isolated RNA from the adherent cells for gene expression through quantitative Real Time PCR; and learned primer designing using Primer-3 program and designed a number of primers related to cancer pathways. She also learned protein expression by Western Blotting (SDS PAGE) and antiplatelet activity. During her short stay at ICCBS she attended the First Nationwide Patch Clamp Workshop hosted by PCMD on December 21-23, 2009. Dr. Thwe Thwe Win gained useful knowledge and skills in advanced technologies related to Molecular Oncology during her affiliation with ICCBS Karachi.
